Acetate Induces Growth Arrest in Colon Cancer Cells Through Modulation of Mitochondrial Function

چکیده

Acetate is one of the main short chain fatty acids produced in colon when fermentable carbohydrates are digested. It has been shown to affect normal metabolism, modulating mitochondrial function, and acid oxidation. Currently, there no clear consensus regarding effects acetate on tumorigenesis cancer metabolism. Here, we investigate metabolic cancer. HT29 HCT116 cell lines were treated with its effect proliferation, reactive oxygen species, density, permeability transition pore, cellular bioenergetics, gene expression acetyl-CoA synthetase 1 ( ACSS1 ) 2 ACSS2 ), lipid levels investigated. was found reduce proliferation both under normoxia as well reducing glycolysis; it also increase consumption ROS levels. Cell death observed independent ACSS1/2 expression. Under hypoxic conditions, reduced maintained line but longer line. together increased hypoxia which may partly protect cells from anti-proliferative reversed Warburg caused by acetate. The findings this study suggest that a consequence impact metabolism during ACCS1/2

Tumor-suppressive miR-34a induces senescence-like growth arrest through modulation of the E2F pathway in human colon cancer cells.

Accumulating evidence suggests a role for microRNAs in human carcinogenesis as novel types of tumor suppressors or oncogenes. However, their precise biological role remains largely elusive. In the present study, we aimed to identify microRNA species involved in the regulation of cell proliferation. Using quantitative RT-PCR analysis, we demonstrated that miR-34a was highly up-regulated in a hum...

Hyperthermic CO2 pneumoperitoneum induces apoptosis in human colon cancer cells through Bax-associated mitochondrial pathway.

Peritoneal carcinomatosis of colorectal cancer is common and associated with poor prognosis, which poses a serious challenge and satisfactory treatments are urgently needed. Hyperthermic CO2 pneumoperitoneum (HT-CO2) is a new strategy. This study was designed to determine the potential of HT-CO2 against colorectal cancer cells. Based on an in vitro HT-CO2 study model, the anti-tumor efficacy of...

Apatinib has anti-tumor effects and induces autophagy in colon cancer cells

Objective(s): Apatinib recently has been used to treat patients with gastric cancer, but the function of apatinib in colon cancer remains unclear. This study was conducted to investigate the impacts of apatinib on the biological function and its potential mechanism of colon cancer cells in vitro. Materials and Methods:The effect of apatinib in colon cancer cells were detected by assessing cell ...

Laminarin induces apoptosis of human colon cancer LOVO cells through a mitochondrial pathway.

Many scientific studies have shown that laminarin has anti-tumor effects, but the anti-tumor mechanism was unclear. The purpose of this study was to investigate the effect of laminarin on the induction of apoptosis in human colon cancer LOVO cells and the molecular mechanism involved. LOVO cells were treated with different concentrations of laminarin at different times. Morphology observations ...

Resveratrol Induces Growth Arrest and Apoptosis through Activation of FOXO Transcription Factors in Prostate Cancer Cells

BACKGROUND Resveratrol, a naturally occurring phytopolyphenol compound, has attracted extensive interest in recent years because of its diverse pharmacological characteristics. Although resveratrol possesses chemopreventive properties against several cancers, the molecular mechanisms by which it inhibits cell growth and induces apoptosis have not been clearly understood. The present study was c...
